Fig. 2

Transactivation of EGFR is involved in the sequestration of Gβγ into the nucleus. A HEK-293 cells expressing D3R (1.8–2.1 pmol/mg protein) were transfected with HA-USP33 and FLAG-arrestin3. The cell lysates were immunoprecipitated with FLAG beads. Co-IP/lysate and IPs were immunoblotted with antibodies against HA and FLAG, respectively. **p < 0.01 compared with other groups (n = 4). B HEK-293 cells expressing D3R (1.9 pmol/mg protein) were transfected with GFP-Gβ1, Gγ2, and arrestin3. The cells were labeled with arrestin2/3 antibodies (1:1000), followed by Alexa 555-conjugated secondary antibodies (1:500). Scale bars represent 10 μm. The Pearson correlation coefficient (γ value) represents the degree of colocalization between DAPI and GFP-Gβ1 (bar graphs on the left) or DAPI and arrestin3 (bar graphs on the right). ***p < 0.001 compared with other groups (n = 5)
