From: RUNX1, FUS, and ELAVL1-induced circPTPN22 promote gastric cancer cell proliferation, migration, and invasion through miR-6788-5p/PAK1 axis-mediated autophagy

Fig. 2

Autophagy can restore the promoting effect of circPTPN22 on GC. A–E circPTPN22 knockdown or overexpression GC cells were treated with autophagy inhibitor 3-Methyladenine or autophagy activator Rapamycin for 24 h after 48 h. The proliferation of GC cells was detected by CCK-8 assay (A), cell colony formation assay (B) and EdU cell proliferation assay (D). B and D are representative clones and fluorescence images, and C and E are quantitative data. Scale bar, 100 μM. F–I circPTPN22 knockdown or overexpression GC cells were treated with autophagy inhibitor 3-Methyladenine or autophagy activator Rapamycin for 24 h after 48 h, and the migration (F) and invasion (G) of GC cells were detected by transwell assay. F and G are representative migration and invasion micrographs, and H and I are quantitative data. Scale bar, 100 μM. *p < 0.05, **p < 0.01, ****p < 0.001
